Suppression from Afar: Striatal D2 Receptors Temper Inhibition in Cortex Turning down Inhibition from Afar

چکیده
11 July 2011. Altering dopamine signaling in one region of the brain can have substantial repercussions elsewhere, according to a study published online on July 5, 2011, in the Proceedings of the National Academy of Sciences. The laboratories of Wen-Ju Gao at Drexel University in Philadelphia, Pennsylvania, and Eric Kandel at Columbia University in New York joined forces to examine the synaptic consequences of overexpressing D2 dopamine receptors in the striatum of mice. They turned up a deficit in GABAergic transmission in the prefrontal cortex—a result that links two disparate findings in schizophrenia and suggests that impaired cognition stems from overactive dopamine signaling in the striatum.

EFFECTS OF CATECHOLAMINES ON DOPAMINE AND SEROTONIN SYNTHESIS IN RAT BRAIN STRIATAL SYNAPTOSOMES: THE ROLE OF PRESYNAPTIC RECEPTORS AND THE SYNAPTOSOMAL REUPTAKE MECHANISM.
The regulation of dopamine and serotonin synthesis in rat brain striatal synaptosomes has been studied using HPLC methods. Noradrenaline was shown to markedly inhibit both the synthesis of dopamine and serotonin. The response of the synaptosomes to the concentrations of noradrenaline appeared to be biphasic, a very effective inhibition occurring at low concentrations (1-5 µm) and a relativ...
متن کاملThe Effect of Inhibition of Dopamine D2 Receptors on Some of the Peripheral Blood Mononuclear Cells of the Rat under Food restriction
Background & Objective: In previous studies, the effects of food restriction on the changes in immune responses and brain dopamine content have been determined. On the other hand, it has been shown that immune cells, in addition to dopamine production, also have dopamine receptors. The purpose of this study was to evaluate the effect of inhibition of D2 dopamine receptors on several functions o...
متن کاملOpposite influences of endogenous dopamine D1 and D2 receptor activation on activity states and electrophysiological properties of striatal neurons: studies combining in vivo intracellular recordings and reverse microdialysis.
The tonic influence of dopamine D1 and D2 receptors on the activity of striatal neurons in vivo was investigated by performing intracellular recordings concurrently with reverse microdialysis in chloral hydrate-anesthetized rats. Striatal neurons were recorded in the vicinity of the microdialysis probe to assess their activity during infusions of artificial CSF (aCSF), the D1 receptor antagonis...
متن کاملStriatal D1- and D2-type dopamine receptors are linked to motor response inhibition in human subjects.
Motor response inhibition is mediated by neural circuits involving dopaminergic transmission; however, the relative contributions of dopaminergic signaling via D1- and D2-type receptors are unclear. Although evidence supports dissociable contributions of D1- and D2-type receptors to response inhibition in rats and associations of D2-type receptors to response inhibition in humans, the relations...
متن کاملDopamine Regulation of Lateral Inhibition between Striatal Neurons Gates the Stimulant Actions of Cocaine
Striatal medium spiny neurons (MSNs) form inhibitory synapses on neighboring striatal neurons through axon collaterals. The functional relevance of this lateral inhibition and its regulation by dopamine remains elusive. We show that synchronized stimulation of collateral transmission from multiple indirect-pathway MSNs (iMSNs) potently inhibits action potentials in direct-pathway MSNs (dMSNs) i...
